Development of an indirect immunofluorescence assay for PCV3 antibody detection based on capsid protein

Abstract Porcine circovirus type 3 (PCV3) is a novel porcine associated with dermatitis and nephritis syndrome (PDNS), reproductive failure, multisystemic inflammation. Capsid protein (Cap) encoded by PCV3 ORF2 gene has been identified as an immunogenic protein. Currently, there no immunofluorescence assay (IFA) available for serological diagnosis. Here, the N-terminal 33 amino acids of Cap were predicted to serve nuclear localization signal (NLS). Two types recombinant plasmids constructed expression in Sf9 cells using baculovirus system: plasmid rvBac-Pc full-length rvBac-Sc honeybee melittin peptide place NLS sequence. Expression sequences was further analyzed IFA. Strong specific fluorescence signals observed nucleus rvBac-Pc-transfected cytoplasm rvBac-Sc-transfected cells. No cross-reactivity 2, pseudorabies virus, classical swine fever or respiratory virus. In summary, we developed two detection modes that can be used detect antibodies. This method suitable diagnosis epidemiological investigation PCV3. study provides reliable monitoring antibody level pigs future.
